I have a John Deere LT 150 150B505432 with a kohler CV15S. 41618 engine that will not start. It has spark and the fuel pump has been replaced. New gas lines new gas filter changed the air filter. Cleaned out the gas tank. New gas gap. Gas lines and gas cap were changed because they were very sticky from the gas over the years. Carburetor was cleaned also. The gas is going to the carburetor and it cranks but will not start. It will not start with starting fluid either. What am I missing?

Sounds like the problem is at the carburetor. It can be a physical or electrical problem.
Verify the fuel solenoid pintle is not stuck and make sure the electrical connector has not spread the pins and not providing power

I have a spark plug tester.How are you checking the spark?
Someone did put an aftermarket carburetor on it. But this carburetor doesn’t have the fuel solenoid there are no wires coming in to hook up.Sounds like the problem is at the carburetor. It can be a physical or electrical problem.

Have you checked the safety switches? Or, are you trying to start it with the blade/pto engaged? Is the parking brake on? If it won’t light off with starting fluid then it has: too weak a spark, a bad spark plug, the timing is off, or the spark is grounding out. Disconnect the kill wire from the coil and see if it lights off. If it does, find the wire that is grounding the spark. Likely to be a safety switch malfunction, or operator error.

What type? Makes a huge difference.I have a spark plug tester.

2.Open, adjustable gap spark tester

Was this new pump tested? New doesn't mean new anymore. Do you have an old cracked pulse hose? Possibly not making a seal? Maybe NO pulse to the pump so she can't pump?fuel pump has been replaced.
