Purchased recently a few new Stihl KM110 combo units, to be used with various attachments.

Once the supplied Stihl oilmix was spent (first gallon), I started using Trufuel 50:1 Non Ethanol premix and the engine(s) start easily but runs kind of rough until I increased both the mix, idle adjustments.

Kind of went by ear, sound, only, is there a better way of doing this ?

The typical way of adjusting two stroke carburetors is to turn the mixture screw until a noticeable change (drop) in R.P.M. is heard then take note of the clock position of the screw. Now turn the screw in the opposite direction and the engine should smooth out and run at a higher R.P.M. but continue turning the screw until the R.P.M. drops again and note the clock position of the mixture screw. The final setting is to turn the screw to the half way point of the two extremes of the lean and rich sides. This is usually a good setting but your particular engine may need fine tuning from there and only you will be able to tell.

Good luck

Slow down, do you have one or two fuel mixture screws? If you have two mixture screws, the previous post will not work. If you can include a few pictures it would help both of us.
